Visualization of Collateral Nerve Fibers of Human Cervical Spinal Cord: Direct Histologic Validation of Diffusion Tensor Imaging

Overview: Line scan diffusion tensor images were obtained in excised human adult spinal cord. The diffusion tensor eigenvalues (11, h2, and h3) and the in-plane angle of the second eigenvector was measured. After the imaging experiments, corresponding histology sections were stained with silver to visualize the collateral nerve fibers and to measure their in-plane angles. Maps of the second eigenvector were overlaid on top of histology images. The angles measured on histology and MRI correlate extremely well (r=0.98). The collateral nerve fibers in human cervical spinal cord can be visualized by mapping the second eigenvector of the diffusion tensor.
